Selective breeding for high and low levels of opiate-induced analgesia in mice.

چکیده
Beginning with a genetically heterogeneous outbred stock of mice (Binghamton HET), selective breeding was conducted within two selected lines for a high analgesic (antinociceptive) response and a low analgesic response, respectively, to a narcotic analgesic (Ievorphanol tartrate) using the hot-plate test. A nonselected control line was also maintained concurrently. Four generations of predominantly mass selection have produced a marked divergence between the two oppositely selected lines, yielding a realized heritability (h 2) o f 0.32 + 0.05. The selection response was markedly asymmetrical, with a realized h 2 in the high direction of 0.45 +_ 0.10 and in the low direction of 0.12 +_ 0.07. These animals represent potentially useful subject material for research concerning the mechanisms and correlates o f opiate-induced analgesia.

Correlation between magnitude and opioid mediation of stress-induced analgesia: individual differences and the effect of selective breeding.
Two experiments were made showing that opioid involvement coincides with the magnitude of stress-induced analgesia. In Experiment I rats subjected to cold water swims were screened for jump threshold levels on electrified grid and divided into high, medium and low threshold responders' groups. Later on the three groups were given 90 s forepaw footshock. Tail-flick latencies rose highest in the ...
متن کاملLow Dose Morphine Enhances Morphine Antinociception Effects in the Animals Pretreated with Selective and Non-Selective Phosphodiestrase Inhibitors
In this study, we investigated the interactive effects of intraperitoneal (i.p.) injections of three different phosphodiesterase inhibitors (PDEIs) on morphine-induced analgesia in mice using tail-flick method. Subcutaneous administration of morphine (1, 3 and 6 mg/kg) caused significant antinociceptive effects in a dose dependent manner. Administration of pentoxifylline (12.5, 25, 50 an...
متن کاملInvolvement of central opiate receptors in modulation of centrally administered oxytocin-induced antinociception
Objective(s): Oxytocin is involved in modulation of many brain-mediated functions. In the present study, we investigated the central effects of oxytocin and its receptor antagonist, atosiban on inflammatory pain. The contribution of opiate receptors was explored using non-selective and selective antagonists. Materials and Methods: The fourth ventricle of the brain of anesthetized rats was impla...
متن کاملOpiate receptor knockout mice define mu receptor roles in endogenous nociceptive responses and morphine-induced analgesia.
Morphine produces analgesia at opiate receptors expressed in nociceptive circuits. mu, delta, and kappa opiate receptor subtypes are expressed in circuits that can modulate nociception and receive inputs from endogenous opioid neuropeptide ligands. The roles played by each receptor subtype in nociceptive processing in drug-free and morphine-treated states have not been clear, however. We produc...
متن کاملResveratrol protects against diet-induced atherosclerosis by reducing low-density lipoprotein cholesterol and inhibiting inflammation in apolipoprotein E-deficient mice
Objective(s):Resveratrol (RES) is a polyphenol compound that has been shown a promising cardioprotective effect. However, some reports have yielded conflicting findings. Herein, we investigated the anti-atherosclerotic effects of RES in apolipoprotein E (apo E)-deficient mice on a high cholesterol diet. Materials and Methods: Firstly, atherosclerosis was induced by feeding a high cholesterol di...
